Historical Study on the Changes of Skin Homagrats in Rats


Abstract
Fifty male and female albino rats were divided into five groups; I. control group, II. total body X-irradiation group (400r, single dose), III. prednisolone administration group (2mg/kg/day), IV. Imur an administration group (5mg/kg/day), and V. splenectomized group.
The prednisolone and Imuran administration were started 7 days before graft and continued until completion of rejection. X-irradiation and splenectomy were done 24 .hours before graft.
Full thickness skin homograft of about 2cmX2cm in size was transplanted on the back of ten rats of each group and the grafts were studied histologically from the 3rd day of operation. The results were as follows.
1. In control group, early neutrophilic infiltration started on the 3rd or 4th day of operation, followed by lymphocytic infiltration on the 4th or 5th day. Vascular proliferation and recanalization was noted from the 4th day, followed by degenerative changes on the 5th or 6th day. The epidermis showed vacuolization of the cells or homogenization from the 5th or 6th day. The grafted junction appeared firmly connected on the 5th day, soon followed by splitting from the 6th day.
2. In total body X-irradiation group, the histologic changes were similar as control group, but the splitting of the grafted junction was delayed until 13th day of operation.
3. In the group of Imuran administration, all the histologic changes described on the control group were delayed, i.e., 3 to 4 days.
4. In the group of prednisolone, all of the histologic changes were similar to those of control group except that the splitting was delayed for 2 to 3 days.
